Responsibilities
Differentiate human iPSCs into intestinal epithelial lineages including 3D organoids, enteroids, and 2D epithelial monolayers
Establish and optimize microphysiological systems to model intestinal barrier function and tissue microenvironments
Develop co-culture systems incorporating immune cells such as macrophages, dendritic cells, and T cells
Design and execute mono-, pairwise-, and multi-drug combination matrices including biologics, small molecules, and pathway-targeted agents
Perform dose-response and time-course experiments under disease-relevant inflammatory conditions
Analyze drug interaction effects including synergy, additivity, and antagonism
Deliver clear, decision-enabling data packages to cross-functional stakeholders
Implement quantitative readouts including TEER, permeability, wound healing, mucus layer, and barrier integrity assays
Perform inflammatory and cell-state profiling using ELISA/MSD, qPCR/RT-qPCR, flow cytometry, high-content imaging, and immunofluorescence
Support transcriptomic profiling (bulk and/or single-cell) in collaboration with data science partners
Apply genetic and pharmacological perturbations to dissect disease mechanisms and predict therapeutic responsiveness
Assess assay reproducibility, robustness, and throughput
Adapt assays to plate-based formats (96-/384-well) and support automation readiness
Partner with Immunology, Pharmacology, and Data Science teams to align experimental models with translational endpoints
Present experimental plans, progress, and results to internal stakeholders and senior leadership
Maintain rigorous documentation, SOP adherence, and data integrity
Ensure compliance with safety practices and proper maintenance of cell lines, reagents, and laboratory equipment
Qualification
Required
Master's degree with 5+ years OR PhD with 1-2+ years of relevant experience in molecular biology, cellular biology, immunology, pharmacology, or related fields
Strong hands-on experience with human cell culture and at least one of the following: iPSC maintenance and differentiation into epithelial or endodermal lineages, Intestinal organoids (enteroids) or epithelial monolayers
Proven ability to design, execute, and troubleshoot functional assays and quantitative dose-response studies
Strong data analysis, documentation, and scientific presentation skills
Excellent verbal and written communication skills in collaborative, cross-functional environments
Preferred
Direct experience with iPSC-derived intestinal models including 3D organoids, 2D monolayers, Transwells, or gut-on-chip platforms
Expertise in barrier function assays (TEER, permeability) and imaging-based analyses
Experience with cytokine and inflammatory profiling
Familiarity with automation, liquid handling, and high-throughput assay development
Background in inflammatory disease biology and preclinical combination testing
